چکیده

In 1939 Schoenheimer, Ratner, and Rittenberg (1) employed nn-tyrosine labeled with heavy nitrogen (N15) to study the metabolism of this amino acid and the fate of its amino nitrogen in rats. The present investigation deals with the metabolism in rats of radioactive nn-tyrosine (tyrosine”), labeled in the /3 position with the long lived Cl* isotope of carbon (2).l The labeling of the carbon skeleton made it possible to measure the incorporation of tyrosine* into tissue proteins and the conversion of this amino acid into other compounds. The moderately high radioactivity of the tyrosine” permitted the administration of single small doses (5 mg.), well within the normal physiological range, and t,he subsequent measurement (with an accuracy of about 5 per cent) of fractions of the order of 0.001 to 0.0001 part of such doses. The present experiments revealed that the carbon chain of the tyrosine, unlike the amino nitrogen, does not contribute significantly to the formation of other amino acids.

Radioactivity distribution in the tissues of mice bearing melanosarcoma after administration of DL-tyrosine labeled with radioactive carbon.

It has long been recognized that radioactive isotopes furnish a means of irradiating deep seated neoplasms and metastases provided they can be administered in a form which will localize with a sufficient degree of selectivity in the neoplastic tissue. The availability of radiocarbon greatly expands the possibilities of this method for controlling cancer. The melanosarcoma suggests itself for lo...

Preparation, distribution, stability and tumor imaging properties of [62Zn] bleomycin complex in normal and tumor-bearing mice

Backgrounds: Bleomycin (BLM) has been labeled with radioisotopes and widely used in therapy and diagnosis. In this study BLM was labeled with [62Zn] zinc chloride for oncologic PET studies. Materials and methods: The complex was obtained at the pH=2 in normal saline at 90 ° C in 60 min. Radio-TLC showed an overall radiochemical yield of 95-97% (radiochemical purity > 97%). Stability of complex ...

Imatinib metabolism and disposition in isolated rat perfused liver

Imatinib is an orally administered tyrosine kinase inhibitor which inhibits the Bcr-Abl protein-tyrosine kinase with high selectivity. Imatinib is rapidly absorbed from the gut, after oral intake and has an almost absolute bioavailability of 98%. The metabolism of imatinib is mediated by the cytochrome P450 (CYP) isoenzymes in the liver and gut wall. CGP74588 is a major active metabolite of ima...

On the metabolism of phenylalanine and tyrosine.

The metabolism of phenylalanine and tyrosine has been the subject of investigation for many years. Much evidence has been produced to show that in mammalian tissue phenylalanine is converted to tyrosine (l-5). Tyrosine can be oxidized to p-hydroxyphenylpyruvic acid, which can in turn form homogentisic acid (5-13). Embden, Salomon, and Schmidt (14) showed that phenylalanine, tyrosine, and homoge...
